Effects of FIS overexpression on cell growth, rRNA synthesis, and ribosome content in Escherichia coli.

چکیده
The Escherichia coli DNA binding protein FIS is a transcriptional modulator involved in the regulation of many cellular processes, including the activation of rRNA synthesis. High-level overproduction of FIS in early, mid, or late log cultures resulted in growth-phase- and media-specific variations in cell growth, rRNA synthesis, and ribosome content. FIS overproduction caused a pronounced increase in rRNA synthesis for late-exponential cultures but a substantial reduction in cell growth and ribosome content. The addition of simple sugars such as glucose or fructose reversed these phenomena, consistent with the functional role of FIS in carbon metabolism.

Proteome analysis of factor for inversion stimulation (Fis) overproduction in Escherichia coli.
The factor-for-inversion stimulation protein (Fis) is a global regulatory protein in Escherichia coli that activates ribosomal RNA (rRNA) transcription by binding to three upstream activation sites of the rRNA promoter and enhances transcription 5- to 10-fold in vivo. Fis overexpression results in different effects on cell growth depending on nutrient conditions. Differential proteome analysis ...
متن کاملControl of ribosome synthesis during the cell division cycles of E. coli and Synechococcus.
The regulation of ribosome synthesis has been investigated for nearly five decades. In earlier studies, the control of rRNA synthesis in bacteria was found to be dependent on nutrient composition of the growth media or cell growth rates, and these observations led to the growth rate-dependent regulation model. Also developed were stringent control, feedback ribosome synthesis, passive regulatio...
متن کاملImplications of rRNA operon copy number and ribosome content in the marine oligotrophic ultramicrobacterium Sphingomonas sp. strain RB2256.
Sphingomonas sp. strain RB2256 is a representative of the dominant class of ultramicrobacteria that are present in marine oligotrophic waters. In this study we examined the rRNA copy number and ribosome content of RB2256 to identify factors that may be associated with the relatively low rate of growth exhibited by the organism. It was found that RB2256 contains a single copy of the rRNA operon,...
متن کاملActivation of Escherichia coli rRNA transcription by FIS during a growth cycle.
rRNA transcription in Escherichia coli is activated by the FIS protein, which binds upstream of rrnp1 promoters and interacts directly with RNA polymerase. Analysis of the contribution of FIS to rrn transcription under changing physiological conditions is complicated by several factors: the wide variation in cellular FIS concentrations with growth conditions, the contributions of several other ...
متن کاملHygromycin B inhibition of protein synthesis and ribosome biogenesis in Escherichia coli.
The aminoglycoside antibiotic hygromycin B was examined in Escherichia coli cells for inhibitory effects on translation and ribosomal-subunit formation. Pulse-chase labeling experiments were performed, which verified lower rates of ribosomal-subunit synthesis in drug-treated cells. Hygromycin B exhibited a concentration-dependent inhibitory effect on viable-cell numbers, growth rate, protein sy...
